make sure 经常是叫别人做什么，祈使句
例：Make sure that you keep your word.你不要食言。
也有用于表决心的时候
例：I'll make sure that I'll never fail the exams.我尽量考试再也不挂红灯。

be sure 经常表达确定肯定的意思，意为某人对某事有把握
例：I'm sure that this dog is male.我确定这只狗是雄性。
Are you sure? 你确定吗？

基本上区别不大，只是一个更注重动作性，make sure更有动作性的意思。
